Adamantane-conjugated Semax peptide with enhanced BBB penetration and prolonged neurotrophic action.

WHAT IS ADAMAX?
Adamax is an adamantyl-conjugated derivative of the Russian **Semax** peptide (ACTH 4-10 analog, sequence Met-Glu-His-Phe-Pro-Gly-Pro). The modification serves two purposes: (1) increased blood-brain barrier permeability via higher lipophilicity, (2) N-terminal peptidase resistance, so intranasal dosing produces ~5-10× longer CNS exposure than plain Semax. Publications from Lomonosov Moscow State University and Bulletin of Experimental Biology and Medicine (PMID 31075176) demonstrate BDNF/NGF-inducer effects and an anxiolytic profile in animal models (forced swim test, elevated plus maze). **No human clinical trials exist**; only user anecdotes and preclinical PK data are available. The adamantyl group recurs in other nootropics (Bromantane, Memantine) – a shared theme of improving CNS permeation.
