WHAT IS CYTIDINE?

Detailed overview

Cytidine is a naturally occurring pyrimidine nucleoside (cytosine plus ribose) that is one of the cleavage products of CDP-choline (citicoline) and is phosphorylated intracellularly to the nucleotides CMP, CDP and CTP for RNA synthesis and for building membrane phospholipids (phosphatidylcholine) via the Kennedy pathway. In humans, however, most ingested cytidine is rapidly deaminated to uridine, so uridine, not cytidine, is the main pyrimidine reaching the human brain (in rodents the balance is reversed). In one randomized trial, cytidine derived from citicoline accelerated the improvement of depressive symptoms in bipolar depression and was associated with reduced brain glutamate/glutamine levels on MRS. Direct human evidence for isolated cytidine is limited: most practical data come from the CDP-choline form.

Mechanism

Pyrimidine and phospholipid precursor (Kennedy pathway)

Human metabolism

Largely converted to uridine in humans

Legal status

Dietary supplement / research compound (not a standalone drug)

Receptor profile

  • Conversion to uridine (pyrimidine pool)Strong
  • Phosphatidylcholine synthesis (Kennedy pathway)Strong
  • Cholinergic / dopaminergic signaling (indirect)Moderate
  • P2Y receptors (via UTP/UDP)Weak

Safety

Side effects, stop signs, contraindications

Side effects · 4

  • Little human safety data exists for isolated cytidine; most experience comes from the CDP-choline (citicoline) form
  • Possible mild gastrointestinal upset (nausea, abdominal discomfort)
  • Theoretical rise in uric acid with large, sustained pyrimidine loading
  • Headache or restlessness can occur, as with many cholinergic precursors

Contraindications · 3

  • Pregnancy and breastfeeding: insufficient safety data for isolated cytidine, avoid
  • Gout or high uric acid: pyrimidine metabolism may theoretically add load
  • Not an approved standalone medicine; the bipolar depression data came under research conditions alongside valproate
