WHAT IS FASORACETAM?

Detailed overview

Fasoracetam (NS-105 / LAM-105 / NFC-1) is originally a Nippon Shinyaku (Japan) development that acts via up-regulation of metabotropic glutamate receptors (mGluR I/II/III). Aevi Medical (formerly Medgenics) advanced it to Phase 2/3 in ADHD (mGluR-mutation-positive adolescents) – initially positive, then mixed results in the 2019 expanded ADHD trial (PMID 31167583). Unique: upregulates GABA-B receptor expression (PMID 10494996) – anecdotally used to support Phenibut withdrawal. Cholinergic HACU enhancement (Coluracetam-like but weaker). cAMP→BDNF signaling cascade for memory formation. Banned in the UK under the Psychoactive Substances Act (since 2016).

  • Headache, often from increased cholinergic activity (mitigated with a choline source such as CDP-choline)
  • Abdominal pain, diarrhea and nausea, mainly at doses above the nootropic range
  • Fatigue, lethargy
  • Irritability
  • Dizziness, mild lightheadedness, especially when starting use

  • Pregnancy and breastfeeding: no safety data, should be avoided
  • Combined with phenibut or other GABA-B-active agents it potentiates their effect (GABA-B upregulation), caution required
  • Prolonged high-dose use may burden liver and kidney function, caution with long-term use
  • Clinical-trial-stage compound, not approved anywhere, long-term safety unknown
  • Not recommended under 18; those on medication or with serious medical conditions should seek medical advice first
