Oxiracetam

Stimulant-leaning racetam, AMPA + cholinergic activity, focus-oriented.

WHAT IS OXIRACETAM?

Detailed overview

Oxiracetam is a 4-hydroxy derivative of Piracetam, generally described as mildly stimulant-like in contrast to Aniracetam`s relaxing profile. Its action combines AMPA-PAM with HACU (high-affinity choline uptake) facilitation, with indirect mTOR-pathway interaction also suggesting synaptic-level structural strengthening (PMID PMC6853072). Memory consolidation, attention, and reaction time improvements documented with chronic dosing and good tolerability (PMID 2518332, 7749739). In demented rodent models, reverses cognitive deficits via neuronal apoptosis regulation. It was marketed in Italy as Neuractiv and Neuromet.

Safety

Side effects, stop signs, contraindications

Side effects · 6

  • Headache from acetylcholine depletion, mitigated by a choline source (Alpha-GPC, CDP-choline)
  • Insomnia and restlessness, especially with late-evening dosing (stimulating-type racetam)
  • Irritability and nervousness at higher doses
  • Mild gastrointestinal discomfort, nausea
  • Mental fatigue or brain fog with insufficient choline intake
  • Dizziness, occasionally brief concentration fluctuations during the initial phase

Contraindications · 5

  • Pregnancy and breastfeeding: insufficient safety data, avoid
  • Severe renal impairment: dose reduction needed, as it is excreted largely unchanged via the kidneys
  • Epilepsy or seizure predisposition: caution due to glutamatergic/AMPA-enhancing activity
  • Concurrent cholinergic agents: combined with a choline source, cholinergic excess (nausea, sweating) may appear instead of headache
  • Stimulants and caffeine: the stimulating effect can stack, increasing anxiety and insomnia
