WHAT IS P7C3?

Detailed overview

P7C3 is an experimental neuroprotective aminopropyl carbazole compound identified by phenotypic screening as an enhancer of adult hippocampal neurogenesis. Its central mechanism is activation of the enzyme NAMPT (nicotinamide phosphoribosyltransferase), the rate-limiting step of the salvage pathway that converts nicotinamide into NAD+, which restores NAD+ levels in injured or stressed neurons and preserves cell survival. In rodent and non-human primate models it showed protective effects in stroke, traumatic brain injury, Parkinson- and ALS-like neurodegeneration, and depression models. There are no human clinical trials: P7C3 exists only as a laboratory research chemical, its safety in humans is unstudied, and it has never been approved for human use.

Safety

Side effects, stop signs, contraindications

Side effects · 4

  • No human safety data: all evidence comes from preclinical (rodent and primate) studies; the compound has never been tested in humans
  • Unknown long-term safety and incomplete toxicity profile in humans
  • Theoretical hepatotoxicity concern was raised for early P7C3 variants; the optimized analogues (P7C3-A20, P7C3-S243) were developed partly to reduce this
  • Systemic consequences of modulating NAD+ metabolism are not characterized in humans

Contraindications · 3

  • Pregnancy and breastfeeding: safety not established, avoid
  • Not an approved medicine; experimental research chemical, not recommended without clinical supervision
  • Known or suspected malignancy: modulating the NAMPT/NAD+ pathway warrants caution in an oncological context
