WHAT IS THN102?

Detailed overview

THN102 is an investigational drug combination developed by Theranexus that pairs modafinil with a low dose of flecainide. Flecainide, otherwise known as a class Ic antiarrhythmic, is used here at a sub-therapeutic dose to inhibit astrocyte gap-junction connexins (Cx30 and Cx43), thereby amplifying modafinil's wake-promoting and pro-cognitive activity. In preclinical models and early human studies (sleep deprivation, Parkinson's disease excessive daytime sleepiness, narcolepsy) the combination showed a more favorable effect on vigilance, working memory and executive function than modafinil alone. It is not approved for any indication and the human data remain limited.

  • Drug interactions: modafinil CYP3A4 induction (contraceptives), flecainide CYP2D6 metabolism (CYP2D6 inhibitors raise its levels)
