WHAT IS VINBURNINE?

Detailed overview

Vinburnine (eburnamonine) is an indole alkaloid of the eburnamine-vincamine family that was marketed in France as Cervoxan for cerebral circulation. As a cerebral vasodilator it widens brain blood vessels, improves circulation and oxygen delivery, and increases the resilience of nerve tissue to low oxygen (hypoxia). Its proposed mechanism involves phosphodiesterase modulation and ion-channel effects, and experimental data also show it allosterically modulates muscarinic acetylcholine receptors. Most evidence comes from preclinical work and older clinical observation: modern large randomized trials are lacking and the compound is not FDA-approved.

Mechanism

Cerebral vasodilation + PDE modulation

Half-life

Short, a few hours (like related vinca alkaloids)

Legal status

Medicine in some markets (Cervoxan), not FDA-approved

Receptor profile

  • Phosphodiesterase (PDE)Moderate
  • Cerebral blood flowModerate
  • Muscarinic acetylcholine receptors (allosteric)Moderate
  • Neuronal metabolism / antihypoxiaWeak

Safety

Side effects, stop signs, contraindications

Side effects · 5

  • Dizziness, particularly on standing due to lowered blood pressure
  • Low blood pressure (hypotension) from the vasodilatory effect
  • Occasional gastrointestinal upset, nausea
  • Rare skin reactions: fixed drug eruption reported in a case report
  • Limited safety data from large clinical trials

Contraindications · 4

  • Pregnancy and breastfeeding: safety not established, avoid
  • Low blood pressure or hypotension tendency: the vasodilatory effect may worsen it
  • Co-administration with other blood-pressure-lowering agents: additive hypotension
  • Known hypersensitivity to vinca alkaloids or the compound
